I can't remember his name, but some guy wrote a book called Bit Literacy that is about cleaning out your mailbox on a daily basis, and how to do it in a way that allows you not to lose any valuable information.

I get antsy and clean out my gmail when it gets to around 200...

I hit inbox zero a while ago when i moved all my mail from my own servers to google apps. It takes a while, but most of my mail was stuff that wasn't relevant anymore, mailing list stuff I could look up elsewhere and any important stuff I filed away in my archive. Even now I'll go through my archive and remove stuff on a fortnightly basis. It's easier than you think... you just have to get used to working out what you really need now.

I also did it at work and I do miss the odd thing now and again, but I have a 2 week period before my trash is emptied on the server-side which is good enough.
I'm a bit overprotective towards my e-mail but I guess cleaning it on a regular basis is probably inevitable, as after some time finding older mails becomes a really challenging task. Still, a couple of hundreds of mails reside in my gmail account :)
